Thermo Fisher's The TaqMan RNA-to-CT 1-Step Kit is designed for RNA target quantification in a one-step real-time RT-PCR format. It combines reverse transcription and PCR in a single workflow, reducing the number of pipetting steps and the opportunities for setup errors. This configuration contains reagents for 2,000 reactions.
The kit has been validated with Applied Biosystems TaqMan Gene Expression Assays and is suitable for a wide variety of RNA targets, including AT-rich, GC-rich and long sequences. Applications stated for the kit include general gene-expression studies, biomarker discovery and microarray validation.
Performance is designed to remain consistent across a broad range of RNA template input quantities. The kit was evaluated across six orders of magnitude of input concentration, supporting quantification over a wide dynamic range. Sensitivity testing with a synthetic RNA template also demonstrated quantification with inputs as low as ten target copies, although results depend on factors such as assay design.
The one-step format provides a streamlined alternative to procedures in which reverse transcription and PCR are performed separately. Together with compatibility with the TaqMan Gene Expression Assay library, this supports straightforward experimental setup for target-specific RNA quantification.
