Thermo Fisher's RevertAid H Minus Reverse Transcriptase - 10,000 U is a recombinant M-MuLV reverse transcriptase for first-strand cDNA synthesis. It has RNA-dependent and DNA-dependent polymerase activity, while a point mutation in the RNase H domain removes RNase H activity. Because RNA in RNA–DNA hybrids is not degraded during first-strand synthesis, the enzyme supports high yields of full-length cDNA from long templates. The enzyme operates optimally at 42–45°C, maintains activity across 42–50°C and remains active up to 55°C. It can generate first-strand cDNA up to 13 kb and can incorporate modified nucleotides, including Cy3-, Cy5-, rhodamine-, aminoallyl- and fluorescein-labelled nucleotides. Applications stated for the enzyme include first-strand cDNA synthesis for RT-PCR and real-time RT-PCR, reverse transcription at elevated temperatures to reduce secondary-structure effects, cDNA synthesis for cloning and expression, generation of labelled cDNA probes for microarrays, DNA labelling and RNA analysis by primer extension. This product is supplied as 10,000 U.
