Thermo Scientific's Phusion HF Buffer Pack contains four 1.5 mL tubes of HF Buffer for use with Phusion DNA Polymerase in high-fidelity amplification. The stated error rate of Phusion DNA Polymerase in HF Buffer is 4.4 × 10⁻⁷. This is lower than the stated error rate of 9.5 × 10⁻⁷ in GC Buffer, so HF Buffer is presented as the default buffer for high-fidelity amplification.
Buffer choice can also depend on template characteristics. GC Buffer may improve performance with some difficult or long templates, including GC-rich DNA and templates with complex secondary structures. For applications in which DNA templates must remain free of detergents, detergent-free reaction buffers are recommended, with microarray and DHPLC given as examples. This product specifically provides the standard HF Buffer formulation in a pack of four 1.5 mL tubes.
