Thermo Scientific's Phusion GC Buffer Pack, Detergent-Free contains four 1.5 mL tubes of detergent-free GC Buffer for use with Phusion DNA Polymerase. GC Buffer is described as a formulation that can improve polymerase performance with certain difficult or long DNA templates, including GC-rich DNA and templates containing complex secondary structures.
The stated polymerase error rate in GC Buffer is 9.5 × 10⁻⁷, compared with 4.4 × 10⁻⁷ in HF Buffer. HF Buffer is therefore presented as the default option for high-fidelity amplification, while GC Buffer provides an alternative for challenging template characteristics. The detergent-free formulation is intended for applications in which DNA templates must remain free of detergents. Microarray and DHPLC are given as examples of applications where detergent-free reaction buffers are recommended. This product supplies four 1.5 mL tubes of the detergent-free GC formulation.
