Thermo Fisher's Maxima Reverse Transcriptase - 10,000 U is an engineered M-MuLV reverse transcriptase developed through in vitro evolution. It has RNA-dependent and DNA-dependent polymerase activity together with RNase H activity. Compared with wild-type M-MuLV reverse transcriptase, the enzyme has improved thermostability, robustness and synthesis rate.
Maxima Reverse Transcriptase supports reproducible cDNA synthesis from 1 pg to 5 µg total RNA at elevated temperatures of 50–65°C. It can generate full-length cDNA up to 20 kb and complete cDNA synthesis in 15–30 minutes. The enzyme remains 90% active after incubation at 50°C for 60 minutes and can incorporate modified nucleotides.
Stated applications include two-step RT-PCR, two-step RT-qPCR, first-strand cDNA synthesis, construction of full-length cDNA libraries, DNA labelling and primer extension. The elevated operating temperature can help transcribe RNA regions with strong secondary structure and can improve specificity when gene-specific primers are used. This product is supplied as 10,000 U.
