Learn more about this product
Thermo Fisher's Maxima H Minus Reverse Transcriptase - 4 x 10,000 U is an engineered M-MuLV reverse transcriptase developed through in vitro evolution. It has RNA-dependent and DNA-dependent polymerase activity, while a mutation in the RNase H domain removes RNase H activity. The enzyme is described as having improved thermostability, robustness, synthesis rate and 50-fold higher processivity than wild-type M-MuLV reverse transcriptase.
The absence of RNase H activity supports synthesis of full-length cDNA up to 20 kb. The enzyme is active up to 65°C, remains 90% active after incubation at 50°C for 60 minutes and can complete cDNA synthesis in 15–30 minutes. It supports reproducible cDNA synthesis from 1 pg to 5 µg total RNA and shows increased resistance to guanidine, formamide and ethanol.
Applications include first-strand cDNA synthesis for RT-PCR and RT-qPCR, reverse transcription at elevated temperatures, cDNA synthesis for cloning and expression, generation of labelled cDNA probes for microarrays and RNA analysis by primer extension. The enzyme can also incorporate modified nucleotides. This product is supplied as 4 x 10,000 U.
