Thermo Scientific's Luminaris Color HiGreen Low ROX qPCR Master Mix - 5,000 x 20 µL rxns is a 2X master mix for qPCR and two-step RT-qPCR using SYBR Green I intercalating dye for DNA detection without sequence-specific probes. It contains Hot Start Taq DNA Polymerase, uracil-DNA glycosylase, dNTPs including dUTP and an optimized PCR buffer with an inert blue dye. The pack supports 5,000 reactions at 20 µL. The formulation contains a low concentration of ROX passive reference dye. It is supplied with 40X Yellow Sample Buffer and nuclease-free water. When the blue master mix and yellow sample buffer are combined, the reaction mixture appears green, providing a visible aid during pipetting. The Hot Start Taq and optimized buffer support specific amplification, while dUTP and UDG provide carry-over contamination control. Template and primers are added separately to prepare the reaction. The master mix supports quantification of genomic, plasmid, viral and cDNA templates. Listed applications include gene expression analysis, siRNA validation, genotyping and pathogen detection, with standard cycling protocols.
